Isolation and characterization of odorous compounds (geosmin and 2-MIB) producing Actinobacteria in North Han River Watershed

Geosmin and 2-methylisoborneol (2-MIB) are organic compounds known to cause earthy and musty odors in drinking water. Actinobacteria can be one of the causes of earthy and musty odors in drinking water. To respond to the odor material produced by Actinobacteria, it is important to specify the distribution and odor-producing potential of Actinobacteria for adequate response to odor causatives. In this study, we have inverstingated the distribution and phylogenetic characterization of Actinobacteria from water, biofilm and sediment in North Han River watershed and then phylogenetic characterization of the isolated Actinobacteria. We isolated 650 actinobacterial colonies and selected 59 representative isolates. The 59 isolates were affiliated to four genera (Streptomyces, Saccharomonospora, Nocardia, Micromonospora) based on 16S rRNA gene sequence analyzed. The potential and expression of odor synthesis genes were analysis by using the PCR method. The genes producing geosmin and 2-MIB were detected in most actinobacterial isolates (88.1%). This study confirmed that Actinobacteria affect the production of geosmin and 2-MIB that causes odors in North Han River Watershed.
